I haven't tried this out yet, but I translated this information from the read me file included with PCEP. (By the way, in case you didn't know, the audio tracks from TurboGraphix/PC Engine CD games can be played in any CD player like normal CDs, so you can convert the audio files to MP3 format just like you would with any regular CD):

The nessecary files:
Audio files:
XX.mp3 (Replace XX with the two digit track number, ie track 1 would be 01.mp3)
Include all audio files in this format. It's possible that not all mp3 formats are supported.
128k would probably be nice and small.
Output is limited to monorual.

Data track files:
XX.iso (Replace XX with the two digit track number. For example, for the common track#2 data track, the file would be 02.iso)
The author of the software used a demo version of DiscJuggler to create these iso files.

TOC file:
XX.toc (Replace XX with any name)
Enter the disk information in a plain text document.
There should be no problem just copying and pasting the data from certain TOC sites. (This is translated from the author's readme file. I don't know anything about these "TOC" sites he mentions. When searching the internet, I did find this site that looks promising though: http://necstasy.emunova.net/)
EXAMPLE: Chouaniki

Track 01 Audio 00:02:00 LBA=000000
Track 02 Data 00:49:65 LBA=003590

Track 04 Audio 02:13:64 LBA=009889

Track 11 Audio 06:46:72 LBA=030372
Track 12 Audio 07:49:15 LBA=035040

Track 25 Audio 21:00:68 LBA=094418
Track 26 Audio 22:02:23 LBA=099023
Track 27 Audio 22:32:51 LBA=101301
Track 28 Audio 24:36:58 LBA=110608
It should look something like this

The files for this CD would be: 01.mp3, 02.iso, 03.mp3, 04.mp3, etc... Plus the .toc file.

Using PCEP's menu, select "toc change" and then select the .toc file. Next try and run the system card.
If you're lucky, it should work.
The author notes that in his tests, Super Darius went as far as the title screen. Tengai Makyo 2 wouldn't even launch. He only made sure that Chouaniki and Castlevania X worked up to a certain point.
