Erase (magic eraser if it's a single colour) the background of the bottom-most layer

Well if it's a .png or something you right click on the eraser icon in the tools area and click either "Background eraser" or "Magic eraser", whichever one works best (magic eraser is kind of like the fill tool, except erases instead of fills). Then with the background eraser choose the size of the eraser at the top, and click and drag the parts of the picture you want to erase. For magic eraser click on the background to erase it. It will be transparent where you erased.

No, no... Goto the blending options and in the first section on the left goto opacity and turn it down a little. That's the real way to do it. Me thinks.

if your making it from scratch just used clear as backgorund. In other words no color what so ever. Or do what chrono mentioned but you have to turn it al the way to 0

You could go to the layer in the Layers box (by default, it's on the bottom right) and adjust the opacity (transparency) using the slider.

If you want a specific object to be transparent and not the whole image, you'll need to seperate your work into layers.

Hope that helps!
